A Study on Protein Enrichment and Digestion Workflows from Multiple Biofluid Types

Mass spectrometry–based proteomic analysis is a powerful tool for biomarker discovery, but the detection of low-abundance proteins in complex biological samples such as plasma, serum, cerebrospinal fluid (CSF), and urine remains challenging due to high dynamic range and matrix-specific workflow constraints. The presence of abundant proteins, such as albumin, can mask lower-abundance analytes, limiting proteome depth and sensitivity for clinically relevant targets, including cytokines and other signaling proteins.


To address these challenges, we introduce the Nanotrap® Protein Enrichment Affinity Kit (Nanotrap® PEAK), which employs magnetic hydrogel particle technology to capture and concentrate low-abundance proteins and peptides,

enabling improved protein detection by LC-MS/MS while simplifying sample processing. With three distinct Nanotrap® Protein Particle chemistries, the workflow enables flexible enrichment across diverse biofluids while maintaining

compatibility with commercially available digestion kits and automated processing platforms.


In this study, we evaluated the performance of the Nanotrap PEAK Discovery Kit Method across multiple human biofluids and assessed integration with commercial digestion kits, including Promega Rapid-Digest, the Pierce In-Solution Tryptic Digestion and Guanidination Kit, the PreOmics® iST Kit, and the Thermo Scientific SMART Digest Trypsin Kit.
