Compatibility with a Downstream Epigenetic Workflow for Colorectal Cancer Detection

In this study, we evaluated three commercial cfDNA isolation kits to gauge their effectiveness in enriching
and isolating cfDNA for colorectal cancer (CRC) detection via epigenetic screening. Methylation-specific real-time
polymerase chain reaction (MSP RT-PCR) and high-resolution melting (HRM) results were used to compare
the Nanotrap® Extraction Advanced Technology Liquid Biopsy Kit (NEAT Kit) to two other commercial kits for
cfDNA extraction. Mock cfDNA samples were created using 1) standard methylated DNA from the HCT116 DKO
cell line and 2) extracted DNA originating from colorectal cancer solid tumor resections; DNA was mechanically
sheared to mimic the fragment size distribution of ctDNA.
