App Note: Nanotrap® Enhancement Reagent 2 Improves Detection of Viral RNA from Wastewater Samples

According to the Centers for Disease Control and Prevention, SARS-CoV-2 can be shed in the feces of individuals with asymptomatic or symptomatic infections, making SARS-CoV-2 RNA detection in sewage a COVID-19 indicator that is independent of healthcare-seeking behaviors and access to clinical testing. Due to low viral concentrations and presence of PCR inhibitors in sewage samples, SARS-CoV-2 viral particles must be concentrated prior to nucleic acid isolation. Common approaches for viral particle concentration include HA filtration, skim milk and PEG precipitation methods. These approaches are time consuming and cumbersome.

Nanotrap® Magnetic Virus particles have already been demonstrated to enable sensitive, rapid, and easy-to-use automated and manual methods for wastewater SARS-CoV-2 testing.

A new reagent from Ceres Nanosciences, Nanotrap® Enhancement Reagent 2 (ER2), improves the binding of viruses to Nanotrap® Magnetic Virus Particles in wastewater samples, thus improving downstream detection of viral RNA. This simple and sensitive method is compatible with column-based RNA extraction kits, such as QIAamp® Viral RNA Mini Kit (QIAGEN Cat# 52906) and AllPrep® PowerViral® DNA/RNA Kit (QIAGEN Cat# 28000), and with bead-based extraction kits, such as ZymoBIOMICS™ MagBead DNA/RNA Kit (Zymo Research Cat# R2135), Maxwell® HT Environmental TNA Kit (Promega Cat# AX9190), and MagAttract® Viral RNA Kit (QIAGEN Cat# 955538).
